Surface Modification for Hemodialysis

We are continuing to work with the Shestopalov lab on surface modifiation of hemodialysis filters to prevent adhesion and activation. Jim recently presented our status and strategy (PDF) to the Technical Development Fund and was awarded the balance of the award,

f-BSA Adsorption on Adarza functionalized pnc-Si chips

Chips of pnc-Si were funtionalized at Adarza. They returned four groups of chips. Ethanolamine, Amino PEG-1, Inactivated sLinker, and Amino PEG-2. These were all incubated with PBS and  5-mg/mL f-BSA  (2:3) in a humid refrigerated dish. The samples were rinsed

f-BSA adsorption on Functionalized pnc-Si (Shestopalov Lab)

Katia in Dr. Shestopalov’s Lab functionalized some pnc-Si windowless chips. The process she follows is shown in Figure 1. She provided chips at various stages of the process. “Grignard”, “Diazirine”, and “PEG”.   After functionalization, the chips were wetted with

f-BSA adsorption pnc-Si

Samples of pnc-Si, 5.5 mm x 5.5 mm, were functionalized at Adarza and incubated with fluorescein conjugated BSA. Wells were formed in PDMS to hole fluid above the samples. Samples were wetted with PBS then f-BSA, 5 mg/mL, was added.

RTP + Silanization (3D format)

As seen before, in 2D format ‘RTP+silanization’ helps. Similarly in 3D format, with membrane side down, the same treatment helps prevent discoloration till 5 days. (samples from W413) Above figure shows how combined effect of RTP and silanization is better

Liquid Silanization again

A bunch of samples from W683 (no RTP) were liquid silanized and used in 3D format to track discoloration in comparison to control sample, in DMEM(-FBS) @ 37C in Oven. I used all the cracked chips which broke while clamping

Silanization and BSA coating

Surface treatments like silanization were seen to slow down discoloration in the past. I carried out liquid amino-silanization (protocol in place) of samples from W683 (no RTP) and used them in both 2D and 3D format to monitor discoloration in

BSA coating on pncSi (3D vs 2D)

I carried out a repetition of BSA coating experiment with both 2D and 3D formats. With 2D, I get the same result as before, but surprisingly, in Sepcon 3D format we observe the opposite to what we see in 2D!

BSA coating on pncSi (2D)

I coated the pncSi samples from W683 (no RTP) with Bovine Serum Albumin (BSA) in Tris-EDTA buffer. Three different concentrations were used: 1 mg/ml, 5 mg/ml and 10 mg/ml (usual range of BSA conc. used for coating Si surfaces). The
